چکیده

HE developmental processes in bacteriophage T4 have been studied extenTsively with temperature-sensitive mutants and amber mutants (EDGAR, DENHARDT and EPSTEIN 1964; EDGAR and LIELAUSIS 1964). In Bacillus subtilis, the sporulation process has been studied with asporogenous mutants ( RYTER, SCHAEFFER and IONESCO 1966; TAKAHASHI 1965). We examined the developmental processes involved in the transition from bacterial spores to vegetative cells with temperature-sensitive ( t s ) mutants of B. subtilis. When incubated at a high temperature (48"C), the mutant spores discontinued their development at various stages of outgrowth. The mutant loci were mapped on the chromosome by transformation and transduction techniques. The results will be discussed from the aspect of sequential expression of genetic information during the course of germination and outgrowth of spores.

Properties of Bacillus subtilis mutants temperature sensitive in germination.

A new mutant of Bacillus subtilis defective in the outgrowth phase of spore germination has been isolated. When incubated at 46 C, the spores of the mutant gave rise to abnormally large swollen cells. Genetic crosses show that the mutant is different from the three previously described. The genetic analysis indicates two regions of the B. subtilis chromosome involved in the control of the spore...

Conditional dihydrostreptomycin resistance in Bacillus subtilis.

Mutants resistant to dihydrostreptomycin were isolated and genetically analyzed in Bacillus subtilis. Two new classes of mutants distinct from the ribosomal strA locus were found. One class, strB, was located between metC3 and ura-1 on the chromosome. The second class, strC, mapped in the spore gene region close to the spoA locus. Both mutant classes were resistant to dihydrostreptomycin during...

Properties of Bacillus megaterium and Bacillus subtilis mutants which lack the protease that degrades small, acid-soluble proteins during spore germination.

During germination of spores of Bacillus species the degradation of the spore's pool of small, acid-soluble proteins (SASP) is initiated by a protease termed GPR, the product of the gpr gene. Bacillus megaterium and B. subtilis mutants with an inactivated gpr gene grew, sporulated, and triggered spore germination as did gpr+ strains. However, SASP degradation was very slow during germination of...

Role of SpoVA proteins in release of dipicolinic acid during germination of Bacillus subtilis spores triggered by dodecylamine or lysozyme.

The release of dipicolinic acid (DPA) during the germination of Bacillus subtilis spores by the cationic surfactant dodecylamine exhibited a pH optimum of approximately 9 and a temperature optimum of 60 degrees C. DPA release during dodecylamine germination of B. subtilis spores with fourfold-elevated levels of the SpoVA proteins that have been suggested to be involved in the release of DPA dur...

Analysis of temporal gene expression during Bacillus subtilis spore germination and outgrowth.

Bacillus subtilis forms dormant spores upon nutrient depletion. Under favorable environmental conditions, the spore breaks its dormancy and resumes growth in a process called spore germination and outgrowth. To elucidate the physiological processes that occur during the transition of the dormant spore to an actively growing vegetative cell, we studied this process in a time-dependent manner by ...
